So just picked up a 10 pack of Nex 500 buffered NAD+ and it appears vacuum has failed. Diluent does not get sucked in when recon'ing. Does this indicate issue w/ stopper seal? Is this ok to use?

EDIT: forgot to add: Google AI says assume it's no longer sterile and discard (replace) so I'm leaning on contacting vendor and returning.

I've never been much of a believer that a vial having vacuum or not has much if any impact on the sterility or safety of the contents.

For those of us who have been doing this for a few years, such vacuum is a relatively recent development. I've spoken to a number of people knowledgeable about the lyophilization process who generally agree that while vacuum is often preferred, it is not a decisive indicator of anything (for instance, vacuum simply not being enabled in the filling equipment during the final sealing) as long as everything was sterile when sealed.

Personally, I wouldn't worry about it.

Thanks - glad I checked here first. I must confess I've taken 3 injections after recon'ing and filtering and it's had the desired effect with no sides. So, I'll keep it 🙂
