FDA has now approved Orforglipron

Not an April fools joke, it would appear.

I'm still waiting for someone to post up "Lilly announces FREE Zepbound Program for all members of the sex work community" with a link to a fake story based around the idea.
Do you need to be an OnlyFans contributor or can any sex worker join?

Beat me to it!

I think a lot of people don’t want to give themselves shots, even if only once per week.

I hope they undercut the sema pill and grab market share.
(Yes I am biased, I now own a little Lilly stock.)

Edit it’s going to be offered world wide (not just US)
And no food/drink restrictions where the Wegovy pill does. I think it’s a good place for them.

"Foundayo"? What image/ lifestyle is that trying to promote/ conjure? At least it's way more fun to say than "orforglipron."

I bet the advertisements will use The Banana Boat song:

Foundayo… Found-ay-ay-o
Daylight come and I feel okay-o

Foundayo… Found-ay-ay-o
Started Foundayo, now the pounds go

Stack that dose and watch it go (Daylight come and I feel okay-o)
Appetite down, metabolism flow (Daylight come and I feel okay-o)

Lift six pounds, seven pounds, eight pounds gone
Feel that energy comin’ on strong
Doctor say, “Hey, you’re doin’ it right”
Scale go down every mornin’, every night

Foundayo… Found-ay-ay-o
Cravings gone and I’m movin’ slow-o

Foundayo… Found-ay-ay-o
Feeling lighter every day though

Snack attacks used to call my name
Now I walk right past without the same
Jeans fit loose, belt notch tight
Confidence comin’ back in sight

Six weeks in and the change is real
Energy up and I love the feel
Friends all ask, “What you been doin’?”
Just smile back like, “Yeah… I’m movin’.”

Etc...

The article mentions that they will match the price of the Wegovy/Sema pill in the States at the lowest dosage for $149 a month. The highest dosage will cost up to $349 a month.

What is the cost of manufacturing a month's supply -- $5.00, maybe. Options are nice. Options people can afford are better. I will get excited when there is competition from more companies.

Good to see this! My guess is this will prompt a lot more interest around here. I think a lot of people will be interested just due to oral compounds being easier to verify safety (don’t have the same sterility, endotoxin concerns)

To reduce GI sides, the official doses are lower than the phase 3 trials:

Gemini said:
PhaseFDA-Approved Schedule (2026)Phase 3 Trial Doses (ATTAIN-1)Purpose / Outcome
Month 10.8 mg (Starting Dose)1 mgInitial "priming" dose to build tolerance.
Month 22.5 mg3 mgFirst titration step.
Month 35.5 mg6 mgLow-range maintenance.
Month 49.0 mg12 mgMid-range maintenance.
Month 514.5 mg24 mgHigh-range therapeutic dose.
Month 6+17.2 mg (Max Dose)36 mg (Max Maintenance)Full Efficacy Dose.

The 17.2 mg dose is considered the "Sweet Spot." It captures roughly 85-90% of the weight loss efficacy of the higher trial doses but with a 40% reduction in reported severe nausea.

I haven't seen any grey/Chinese tablets below 6 mg, and I only see 12 mg usually. Average cost is about $200 for a hundred 12-mg tablets (14 to 19 cents per mg). They can be cut, but they are small to begin with (at least with Uther).

For me as a hoarder, the biggest problem is the tablets may not last as long as kits, with Uther saying three years before expiration for the tablets. And grey pricing is not exciting so far and hasn't really changed since starting last fall or so.

The main benefit for stacking or aggressive titration is the shorter half life (24 to 36 hours for most/fast metabolizers). But EOD (every other day) dosing may be best to prevent sides, if not splitting the tablets. Third or forth day can be rougher than the first, from what I remember taking it.

I would not put stock in the three year thing. Almost all dry pills stay good years after "expiration".

True, thanks. I could also vacuum seal my second pill bottle, if I don't plan on using it this decade. But I will probably use them all sooner than that, like as a shorter-term sema for stacking.

The Phase 2 trials showed that the most effective dose for weight loss was actually 24mg: https://www.thelancet.com/cms/10.10...c57-8753-d2b4ce7c13c9/main.assets/gr2_lrg.jpg

I realise a lot of people are not comfortable with injecting themselves, but a near painless injection once a week for more weight loss and less side effects for tirzepatide ( or reta ) would seem to be a better option than any of the oral GLP's. Putting up with more nausea or vomiting does not seem like a great trade off to avoid an injection.

I do appreciate the short (relative to the popular injectables) half-life. If someone desired to cycle off having their receptors activated at supraphysiological levels 24/7 for a week or two periodically, it would enable a person to more easily do that (VS the 2-3 months that the injectables would typically take).

To be clear that's not some misguided receptor desensitization notion, but simply an observation that there may be bodily systems that only activate when GLP-1 signaling is low and perhaps having those systems forever suppressed may come with a cost that we're not aware of.

While this may be different The US army under the Shel Life Extention Programme have tested solid medicications that are years past the expiration date and they have been actually pretty good

Obviously things may be different, but usually manufacturers put a large margin for error for expiration dates. It's a risk, but it's up to everyone to check the risk

Interesting that there is still noone reliably selling this. I have heard that there issues trying to recreate it properly.

You won't find orfo on most pricelists.
But it is available both domestically and internationally from a few select vendors.

I got it twice, once tablet, once liquid. Neither are available anymore. Worked really well though, hope to see it come back in a reputable way.
