Hi all,

I hope someone can help me with this :)
I used freezedried Sema 10mg vials for quite some time and dilluted it with NaCL 0.9%. Always kept it in the fridge and solution was okay for > 2 months.

Now I bought raw semaglutide powder frome same distributor. Dilluted it with NaCl and added 1% of pure benzylalcohol (by myself) and than filtered it with .22 micronfilter and made my 10mg in 2 ml vial.

Tried it multiple times, but always after 2 or 3 weeks the solution gets flakey, and strings will form in the liquid. After a little bit of shaking, the flakes and strings go away, but will form again after 30 min. What is going wrong? Why is the solution not stable? Do you have any tips for this? Is is degrading faster?

Could it be the because of the pH of the filling agent? Should I try ‘normal’ 0.9 NaCL? Or BAC water of the shelf?

Hope someone can help me with this. Thanks in advance.
Best regards!

Sorry I don't get it, you buy raw Sema in bulk that's not lyophilized? Why do you do that? You can get a year supply for like 150$

Of course raw isn't stable and isn't meant to be used that way at all, vendors don't just do it for fun, there's a good reason why it's lyophilized with fillers and splitted into vacuum vials
