Do you wet your filter?

I had watched the peptide testers videos about filtering a few times, but recently saw a video on STG in which the person put "diluvent" into the filter first
to wet it and let it sit while prepping everything else. Then used it to filter the peptides she was combining into a vial.
Have any of you heard of this step in the process, is it helpful or advisable??
I have had challenges with filters clogging and breaking, so I am wondering if wetting them first is a good idea?
 
can't you wet it with bac ?

draw 3ml of bac in a syringe

inject 1ml bac into peptide vial
inject 1ml bac in donor vial through the filter

draw recon peptide in another syringe and inject through filter

inject last ml of bac to flush the filter

Good question I just bought filters. It was hard squeezing it thru.

Just try to take your time with the plunger. I use a 25Ga 1½" needle on my 3ml luer lock barrel for recon/extraction (gets to the bottom of the vial). Once my reconned peptide is in the barrel (with filter affixed) I use a 27Ga ½ needle on the filter to dispense into final destination vial. The original 1½" needle i use as the vent.

Yeah my venting filter needle didn't work on a vial and I lost 70mg of glow
Same as above and make sure your vent needle barely enters the vial so you don't get back flow or foaming out the vent.
 
I used the 5/8 for venting multiple vials, I guess it got blunted by the time I got to the pen cartridge

Now I just filter the final cartridge like in videos not the bac and the puc vial

It's possible a bit of stopper clogged your vent. If used multiple times you raise that possibility each time.

yes I assume they said diluvent for those who use sterlie water etc..
 
Air didn't move out through the air filter needle

I pushed glow in through the other needle filter

Rubber plunger got pushed out and it leaked
 
The only reason I can think to 'wet' a filter is to fill, prime, the void inside it with a liquid. When dry, the filter will retain 5-15 units of the solution (like a sponge) which can skew your calculations. I suggest you practice filtering using bac water first, on a old vial. I never had filters breaking or clogging in one year. I use PES 0.22um 13mm.

The first few times it went completely smoothly. I use the 4mm ones, and living in Hawaii, these things cost me much more than anyone here, so I am kind of frugal with them.
My mistake was kindly pointed out to me by my friends here; I messed up in not waiting long enough for the peptides to settle and dissolve. I assumed 10 mins would be enough.
I think the drawing and filtering bac into the vial first is a good idea, both to fill the void and to wet the filter if that may be helpful.
I need to recon some peps soon so I want to get it right this time.
