When using peptides. Does everyone just...

Not really sure the point of what you are asking so I will give my entire process from start to finish.

I have 10mg vials of semaglutide. I store them in the freezer.
I take out a vial to warm up for 10 minutes or so.
I reconstitute with 2ml of BAC water.
I transfer the contents of the vial into a injection pen cartridge.
I put the cartridge in the pen. (photo of pen assembly below)
Remove excess air from the cartridge.
To do this I waste an needle and dial in "doses" with the pen and hit the button until the air is all gone. I'm sure you could find a youtube video of the process if you are curious.

My dose of sema is 1 mg per week.
I have used a peptide calculator and it tells me that I need 20 units.
On my injection day I attach a needle, set the dial to 20
I jab myself in a squishy spot and push the button.
Remove the needle and dispose of it safely.
When my pen is not in use I keep in it a pencil case in the fridge. I do not pay any attention to temperatures.

There are two things here that I admit are problematic.
#1 - I don't filter. Doing this increases the level of risk I am taking.
#2 - I keep and use reconstituted vials for more than 28 days.

I might start filtering at some point. But as it is now I haven't looked into the process and I don't have the supplies. I will continue to use cartridges for longer periods of time. I don't personally believe that they expire that fast. But that is solely my own opinion with no medical facts to back me up. If it ever becomes a problem then I will only have myself to blame.

When I take my Dupixant, I let it sit out, it’s a lot of liquid, I let it sit for hours sometimes. When I do peptides I fill, clean up the wrappers, paper towels and alcohol pads, then inject.
Dupixant is safe sitting out for days though(14 days before it’s bad), not sure about peptides.
 
The amount drawn into the syringe is so tiny it's going to be close to room temperature by the time you inject it.

LET'S DO THE MATH

Well, let's ask an AI to do the math, because I am lazy and I am not going to look anything up. I did measure a 1 cc insulin syringe at 2.5 g though, and I think it's made from polypropylene. So if we assume the syringe is at 68F and the liquid in the vial is at 40F... When the liquid comes to equilibrium with the warmer syringe, it will rise to about 62F.

That's obviously a highly simplified model but ...

Just inject it.

I don't see the point of waiting, what could be the benefit?
 
I asked because I've had some really crappy nights when taking tesa. Terrible sleep/insomnia. Only on .4mg and ONLY happened on the first pin of a new vial. But EACH new vial was the same crap night. No issues after initial pin. I was a bit concerned so called and the pa who I got the stuff from suggested that I let it sit out for 20 minutes before using. I've since went to 1mg a couple of weeks ago no issues-no new vial YET either so there's that.
Anyway I was recommended for the tesa because I initially thought trt might explain a couple issues ( one of them lousy sleep ) however blood work came back at 860 so she couldn't prescribe test. She laughed a little when she said it ( I'm coming up fast on 70 so it is kinda funny ) and obviously I don't NEED it though I still kinda wanna try it. 😬
Switched to Reta from sema about 3 weeks ago. Started Sema 7/11. My first venture with peptides and it was really just to try to lose "stubborn belly fat". I was mostly carnivore for over a year but hit plateaus for 6-8 months and grew frustrated with the stall. Have dropped 20lbs ( CW 179lbs ) or so since July so I'm satisfied, mostly, with this. Pinning 2mg reta ( .5 to start and then Img for a week and am at 2mg now-starting yesterday and then Thursday and intend on 3mg Mon/Thur next week. Unless I have side-effects people talk about.
So that's why I asked. Tonight I will go straight from the fridge.

Could the pa have meant, let it sit for 20 mins after reconstituting, to make sure it's fully dissolved?
 
2 main reasons for letting a peptide bottle come to room temp: the peptide might have precipitated (solidify in tiny amounts) and allowing it to come to room temp allows it to re-dissolve; any condensation that could lead to contamination (on the syringe) can be wiped off.

No. It was understood that it had been reconstituted and had been refrigerated and was cold... and knew that it had occurred with 2nd vial as well.
As I had said I would yesterday I took it straight from the fridge-no issues.
Just weird that it only happens on a new vial. I have one left ( from ReviveRx ) of the three that I started Tesa with. I'm using a different sellers vials at the moment ( and haven't had the "first pin" problem ).
I have another scheduled telecall Friday.

I would expect that it would be every pin and not just the first one if your premise was true. They may have thought that that had occurred. My experience eliminates it as a possibility. And he also suggested maybe take in the mornings. I decided to try it at night again just to see if it would duplicate the sleeplessness but nope, just that first shot. I did let every one of 'em sit out for 20-30 minutes for weeks but after last night I think I'm good for straight from the fridge. I'll continue that unless there's a problem that arises.
I do suspect that last ReviveRX 12mg vial will give me problems on the first pin. 😬

This is really interesting. Try turning and rolling the vial around before drawing the first time.
It sounds like a compounding pharmacy product, and they may add a substance that sinks or floats, which you draw up more of the first time, depending on how you are doing that. This would explain why it affects you just the first time. What else is listed in their ingredient list?

Heck if I know what else is listed...at the moment, anyway. Wait. I'll get vial...
12mg Tesamorelin acetate, 60mg Trehalose, benzyl alcohol ( preservative )

That answer anything for you?
