Orforglipron: The next big thing?

Novo Nordisk is working on a pill form of Semaglutide using their "SNAC" technology. The recent news about Amycretin was also encouraging, although still in early trials. I agree that pills will be the preferred method of administration for most people. This would mean easier production, distribution and storage. Unfortunately, I am less optimistic that Novo or Evil Lilly would charge any less for the pills. We need competition from more companies, not just a different form of the medication.

If the drug costs less to make , store , transport and distribute the price will be lower. More so if there are multiple oral drugs on the market. In fact they may be able to make MORE money charging less. The vast majority of Americans aren’t able to afford the current price structure. 50% profit levels on 10 sales are a lot better than 200% profit levels on 1.


There are 104 million obese Americans. And 86 million overweight. Around 15 million are on glp 1 drugs ( legit scripts ) currently. The drug companies could sell a LOT more at a lower price point. And they are going to have to in a few years when patents expire.

Agreed. Over time the prices will fall. Granted I’m not paying much now thanks to Chinese vendors.
 
No thanks to a pill form of anything. I have to think about my tirz once a week for about five minutes, and then I get to just go about my business. If I could stick my thyroid medicine, my vitamins, and all my other meds into a syringe, do it once a week, and spend the whole rest of the week never trying to remember to go swallow a pill, I'd be thrilled.

I'm not sure I actually prefer an injection to pills, however, I don't mind injecting myself enough to tolerate worse results with pills. I do realize that large numbers of folks do have problems with injecting themselves. I though I had a problem with injecting myself too as I spent about an hour preparing to inject myself for the first time. Then I injected myself. It was actually rather funny; after all that worry, it turned out to be nothing (for me).

GIRL SAME! (or boy same) SO SAME! I thought it was gonna hurt, gonna be scary, gonna be complicated, and then "Oh. Now I'm done? for a WEEK???"
 
I had nightmares about stabbing myself after my first injection. I kinda like pinning myself now. I also had nightmares when I first got contacts. I still don't really like putting those in and taking them out.
 
I noticed they have Tirz in pill form. Anyone have info on this? is it effective? Has anyone given it to their rats yet?
Oral tirz is a scam. Stomach acid destroys peptides. NN has an oral formulation of semaglutide but it requires a special formulation and very high dose to make it through the stomach intact

I feel it would be such a silly thing to even try. Tirz is once a week and works incredibly well.

I totally agree. But I have a needlephobic friend that i'm thinking about. Orfor is only a GLP-1 while Tirz is dual with GIP was my thinking. But so far, the consensus is that a Tirz in pill form won't work.

If your friend is brave enough there is always being a test subject for bioglutide.

I thought this might be useful for discussion. Although since these are different studies with varying timelines its not exactly as it looks. The depth should be % weight lost of the time specified above each section. This I took from a sub reddit, posted by a mod who I believe is associated with Pharma.

Well they could always try and make a GLP1 suppository. ROFL.
 
I estimated where each of the leaders weight loss percentage would have gotten to, if extended to 48 wks like the reta study, and drew lines across for comparison. The only ones that come close to Reta are the Cagri-sema and the Mazdutide..

Thanks. It seems like the best bet for next generation weight loss drugs really got now is Reta. For FDA approved stuff, tirzepatide is the clear winner for most folks. I have had to stop because I’m having liver problems. While the doctor thinks reta is probably not the cause, I have to stay off reta until my liver results come back to normal.
 
I'm sorry to hear about that, and I'm thinking we should all get tested regularly. I havent in a year.
